Biochemical Mechanism of Platelet Aggregation Induced by Nitrogen


Abstract
Nitrogen, as major etiology of decompression sickness, is associated with platelet aggregation in several molecular moiets. Soluble nitrogen act as a platelet agonist, but biochemical mechanism are not well known.
Since the property of a platelet agonist is to potentiate the response by another agonist in a synergistic manner, combination test between nitrogen and the classical agonist was performed.
Epinephrine, showed irreversible aggregation profile, and with nitrogen caused a strong, synergistic effect: This synergism was only partially inhibited by indomethacin and ASA. ADP and nitrogen had different aggregation profile: ADP only show
reversible aggregation profile, and combination with nitrogen cause the reversible ADP aggregation irreversible.
This result may suggest nitrogen induce platelet not by secondary effect, mobilization of classical physiologic agonist, but acting as an independent agonist, different from the well-known "classical agonist."
